832 3D Bioprinted vascularized human skin models can be used to screen modulators of angiogenesis for medical and dermo-cosmetic applications

Lack of skin vascularization or in contrast its hypervascularization are aggravating factors some medical pathologies (ulcers and cancers) dermo-cosmetics troubles (rosacea dark circles). Thus, developing a suitable vitro model to screen pharmaceutical cosmetics ingredients able stimulate mitigate the angiogenesis process without changing normal physiology not easy hampered by existence unreliable models. Traditional models concentrate on analyzing co-culture growth endothelial cells with added fibroblasts keratinocytes. However, these do work for topical applications top-down layering analysis required complex studies translate well clinical analysis. Here we describe creation 3D bioprinted vascularized model, designed screening those type molecules. Human Dermal Microvascular Endothelial Cells, dermal epidermal keratinocytes were harvested expanded from juvenile foreskins. Cells selected optimal mixed bioink into which adhesion proteins cartridged CELLINK pneumatic bioprinting system allowing generation printed full thickness skins. Resulting had bed created lower quadrant structure, was confocal microscopy imaged anti-CD31 fluorescent labelling. Reduction factor pituitary supplement resulted sequential reduction cell death vascular over 14 days, whilst continual supplementation maintains cellular interconnections. With Bioprinting can produce hundreds one afternoon giving potential many combinations molecules before moving more expensive experimentations.
